Caracterización de la respuesta inmune generada por la inmunización e inmunoterapia de una proteína antigénica de trichinella spiralis por Chávez Guajardo, Elsa Gabriela

“…Así se plantea como objetivo de este estudio caracterizar la respuesta inmune generada por la inmunización e inmunoterapia de la proteína de 45 kDa y del Antígeno Soluble Total (AST) de Trichinella spiralis, para el estudio se utilizaron ratas cepa Long Evans, divididas en cuatro grupos, control sano, control infectado, grupo inmunización y grupo inmunoterapia estos grupos tanto para hembras y machos y para proteína de 45 kDa y AST, para la inmunización se realizó una aplicación semanal de la proteína correspondiente vía intradérmica, subcutánea, intramuscular e intraperiotoneal durante cuatro semanas, posteriormente se retó con T. spiralis, se tomó muestra sanguínea de cada animal cada dos semanas por 6 semanas para aplicar las técnicas indirectas, posteriormente se sacrificaron se obtuvo muestra de musculo esquelético para la aplicación de técnicas directas, para la inmunoterapia primero se infectaron los animales y se monitorearon hasta completar el ciclo vital del parasito, posteriormente se aplicó la proteína correspondiente 5 aplicaciones, subcutánea en días consecutivos y concentración ascendente, se realizó la purificación del AST para someterlo análisis por espectrometría de masas, los resultados de las técnicas directas muestra modificaciones a las LI obtenidas por las distintas técnicas, también se obtuvo disminución de la carga parasitaria con respecto al control infectado, al realizar un análisis de varianza para los datos obtenidos la razón de varianza indica que existe diferencia significativa entre el control y los tratamientos, el Wester Blot mostro un patrón de bandeo donde se identifica el reconocimiento de la banda de 45 kDa y el tratamiento que muestra esta banda desde las segunda semanas y hasta la sexta semana, los cortes histológicos sometidos a tinción Hematoxilina-Eosina muestran un importante infiltrado celular de células polimorfonucleares, esta reacción se manifiesta exacerbada en muestras procedentes del tratamiento de inmunización con la proteína de 45 kDa, estos resultados muestran que la inmunización con la proteína de 45 kDa confiere protección contra la infección por T. spiralis. …”
