“…Therefore, wt1 is a good target for the development of gene therapy strategies and its silencing could have a dual benefit, first reducing tumor cell proliferation and second, inhibiting tumor angiogenesis. In this work we investigated the potential of wt1 as a therapeutic target by exposing the murine melanoma line B16F10 to a complex of RNAi against wt1 (RNAi-wt1) in combination with chemotherapeutic drugs (Gemcitabine and Cisplatin), where a synergistic effect was determine by a isobologram; besides, a clonogenic test was performed, obtaining a decrease in the number of cell foci; in the PCR techniques a decrease in the expression of wt1 and the apoptotic gene bcl2 was observed; and in the cytometric test, a reduction in cell viability by apoptosis of 22% was found. …”
